dependence is marked first by an increasedtolerance, with more and more of the substancerequired to produce the desired effect, and then bythe appearance of unpleasant withdrawalsymptoms when the substance is discontinued.
譯文：
對藥品的依賴性首先表現為不斷增長的耐藥量，要產生理想的效果所需的藥品劑量越來越大，而一旦中斷使用，就會出現痛苦的停藥癥狀。
點睛：
句子的主干結構是dependence is marked firstby…，and then by...。逗號隔開的成分是with復合結構，with+名詞+過去分詞，作伴隨狀語，進一步解釋藥物依賴的第一種表現。when the substance isdiscontinued是時間狀語從句。
另外注意名詞在這里的特殊意義：dependence指“對藥物的依賴，毒癮”，withdrawal的意思是“停止用藥”。
